Description: Mesothelin (MSLN) is a glycosylphosphatidylinositol-linked membrane protein that is expressed on mesothelial cells lining the pleura, pericardium, and peritoneum. The gene encodes a 71 kDa precursor protein that is processed to yield a 31 kDa shed protein called megakaryocyte potentiating factor (MPF) and a 40 kDa protein called Mesothelin which is attached to the cell membrane via a glycosylphosphatidylinositol anchor. Although the biological functions are unclear, several studies have demonstrated that overexpression of MSLN is involved in cell adhesion and tumor invasion. Overexpression of MSLN has been observed in several cancers such as mesothelioma, ovarian cancer, pancreatic and lung adenocarcinoma. Due to its high expression in tumors, MSLN could serve as a potential target for anti-cancer therapy. Current strategies involve developing CAR-T therapy and monoclonal antibodies that target MSLN. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. HRP-Streptavidin is added to the plate, and post-incubation any unattached conjugates are washed off by Wash Buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of MSLN in the sample or standard.
