Description                       The antiserum represents an excellent marker to study terminal epidermal differentiation. The ab is reactive with epidermal cells in uppermost suprabasal layers including scalp, foot and sole. It recognizes individual cells within epidermis of tongue and mamille (co-localization with cytokeratin 1). It is also strongly reactive on hyperkeratosis of diverse origin.

Tissues Specifically Detected The antiserum represents an excellent marker to study palmoplantar epidermal distribution and differentiation, specifically reactive in the middle/upper suprabasal layers (stratum spinosum/ granulosum) of the epidermis of palm and sole. CK9 can be detected in primary cultures of palmoplantar keratinocytes when they shift to differentiation-promoting conditions and grow stratified (upper cells). CK9 has not been found in normal, i.e. non-pathogenic, non-ridged epidermis, beside some minor cells surrounding the acrosyringeal ducts. No labelling has been found in epithelial cells of other stratified epithelia such as oesophagus or complex epithelia (e.g. urothelium) or in ductal or simple epithelia. Negative tissues include: muscle, liver and duodenum. (see references listed below).

Reactive Polypeptide

(Specificity) MW 50,000 0(pI4.9) intermediate filament polypeptide, cytokeratin 15, detected by immunofluorescence microscopy in basal cells of stratified epithelia (including epidermis, tongue, esophagus, exocervix) and complex stratified epithelia (eg bronchial epithelium), in luminal epithelia of mammary, salivary and sweat gland. In the hair follicule, staining was restricted to the outermost cell layer of the outer root sheath; within the hair follicule several cell layers of the hair follicule were positive with antiserum gp15.1.

In human fetal tissues, cytokeratin 15 was localized in basal cells of epidermis, lung, trachea, tracheal glands and esophagus. In fetal hair anlagen, all cells were positive with antiserum gp15.1. The antiserum is completely unreactive with cells of liver, colon, urinary bladder, pancreas and lung alveolus.

In tumors, gp15.1 is reactive with squamous skin and lung carcinomas and with a significant subgroup of invasive ductal breast carcinomas, especially tumors with higher grade malignancy (Heid, Bartek, Leube, Moll, Kaufmann & Franke, unpublished results)
