In western blot, this clone reacts with the TC(b) fragment of pepsin digested type I collagen after digestion with mammalian collagenase. It also reacts with lathyritic type II collagen. The binding epitope is localized to the carboxyl-terminal one quarter of hte type II collagen. It recognizes the ai(II) chain but not the a3(XI) chain, The epitope for clone 2B1.5 Mab is not susceptible to digestion of lathyritic type II collagen with recombinant stromelysin.

The epitope for clone 6B3 is localized in the triple helix of type II collagen. Immunoblotting of CnBr peptides of collagen II shows that clone 6B3 Mab reacts with CB 11 fragment which is the site of immunohgenic and arthritogenic epitopes alon the intact II molecule. In western blot (but not in Elisa) this Mab recognizes a1(II) as well as a3(Xi) chain both of which have identical primary structure.

The epitope for this Mab is lost after digestion of lathyritic type II collagen with recombinant stromelysin.

Localization: Basement membrane
Background: Collagen IV is a major constituent of the basement membranes along with laminins and enactins. It is composed of � 1(IV) chain and � 2(IV) chain in 2:1 ratio . It can form insoluble fibers with high tensile strength. Antibody to collagen IV is useful in detecting the loss of parts of basement membrane in carcinomas. This MAB cocktail is highly specific to type IV collagen. In kidney, it reacts with glomerular and tubular basement membranes, parts of mesangial matrix and the Bowman�s capsule. It also reacts with basal lamina of capillaries as well as basement membranes in a variety of tissues. This Mab cocktail is excellent for the staining of routine formalin-fixed, paraffin-embedded tissues.

Clinical Application/Specificity The antibody is reactive with collagen type IV of basement membranes, and shows a homogeneous staining pattern in all tissues. As neoplastic cells of invasive carcinomas often lack a continuous basement membrane, the antibody is useful to distinguish between non-invasive and invasive lesions. Additionally, it can be used for the differentiation of bullous lesions in dermatopathology. No cross reactivity with other collagens at optimal dilutions. In immunoblotting1, a slight cross reactivity with collagen type V is observed.
