Reactivity: The monoclonal antibody is directed against the the CD32-antigen,which is expressed on granulocytes,B-cells, eosinophils and macrophages (1,2). The CD32-antigen is the FcR-II receptor for aggregated IgG, and it has also a function in cell activation.The presence of CD32-antigen has prognostic significance in patients presenting with AML.Acute leukaemias with a high proportion of cell bearing the CD32-antigen may be similar in prognosis to the chronis forms of leukaemia (3,4).
Application: Diagnosis of acute leukaemias by flow cytometry.
Immunoperoxidase staining of frozen tissue sections
Methods: Indirect immunofluorescence staining with analysis by FACS or fluorescence microscopy.

Activity: Reacts with the 40KD type I membrane glycoprotein, FCgammaRII ) , the low affinity receptor for aggregated IgG expressed on B cells, monocytes, granulocytes and platelets. CDw32 mediates endocytosis, cytotoxicity, platelet activation and immunomodulation. Studies indicate this clone binds with FCgammaIIa, IIb1 and IIb1*.

Immunogen: Human K562 tumor cells and L cells transfected with human Fc gamma RII
PRODUCTION: Antibody was Protein A purified from (low FBS containing) tissue culture supernatant. Immunoglobulin was enzymaticly cleaved using Immobilized Ficin. F(ab')2 was separated from intact antibody and Fc fragments by protein A and Size exclusion chromatography. Purity was >90% F(ab')2 by SDS-PAGE.
Activity: Antibody 7.3 reacts with a domain 2 epitope or all CD32 isoforms. Intact and Fab'2 7.3 antibody blocks immune complex binding.
STORAGE : Store at 2 - 5oC. Open under aseptic conditions. Freeze/Thawing is not recommended.
INFORMATION: Human CD32 exists in at least six isoforms originating from three different genes (FcgRII A, B and C). The CD32 molecule is a low affinity receptor for immune complexed IgG and has signal-transducing capabilities involved with humoral and cell-mediated immune responses.

PERFORMANCE: Five x 10 5 cultured human Raji cells were washed and incubated 45 minutes on ice with 80 ul of anti-CD32 F(ab')2 at 20 ug/ml. Cells were washed twice and incubated with 2o reagent Goat anti-Mouse IgG/FITC , after which they were washed three times, fixed and analyzed using a BD FACS Calibur. Cells stained positive with a mean shift of 1.32 log10 fluorescent units when compared to a Mouse IgG1 negative control at a similar concentration.
