MOUSE ANTI-HUMAN CD106/VCAM-1 MONOCLONAL ANTIBODY
cat# RDI-CD106abm-B1 $312.00
Presentation: 100ug in 0.1ml (1mg/ml) 50 mM Sodium Phosphate pH 7.5, 100 mM Potassium Chloride, 150mM NaCl, .5 mg/ml Gentamicin Sulfate (as a preservative).
Clone: 1.G11B1
PRODUCTION: Antibody was Protein A purified from (low FBS containing) tissue culture supernatant. Purity was >95% Immunoglobulin by SDS-PAGE with less than 1% Bovine Immunoglobulin.
Isotype: Mouse IgG1
Immunogen: Human endothelial cells
INFORMATION: Human CD106 is an endothelial adhesion molecule that binds to a4b1 & a4b7 integrins(VLA-4) and promotes adhesion of lymphocytes, monocytes, eosinophils and basophils. CD106 is also found on follicular dendritic cells, interdigitating recticulum cells and Kupffer cells. Antibody 1.G11B1 recognizes the VCAM-1 molecule (CD106) and blocks leukocyte adhesion.

STORAGE: Store at 2 - 5oC. Freeze/Thawing is not recommended.
PERFORMANCE: Cultured human umbilical cord vein endothelial cells were stimulated 4 hours with 10 ng/ml Phorbol 12-Myristate 13-Acetate. Cells were washed and incubated 45 minutes on ice with 80 ul of anti-CD106 antibody at 5 mg/ml. Cells were washed twice and incubated with 2o reagent Goat anti-Mouse IgG/FITC at a 1:60 dilution factor, after which they were washed three times, fixed and analyzed using a BD FACS Calibur. Cells stained positive with a mean shift of 0.89 log10 fluorescent units when compared to a Mouse IgG1 negative control (Catalog#RDI-MIGG1X1, $188.00) at a similar concentration.

SPECIFICITY: The antibody is specific to human vascular cell adhesion molecule (VCAM-1, 110 kDa) whose ligand is VLA-4 (CD49d/CD29, a-4/b-1 integrin subunits). The antigen is distributed across non leucocyte and leucocyte cells e.g. neural crest and fibroblasts, B cells, T cells, monocytes and large granular lymphocytes
APPLICATIONS
*This antibody works both in ELISA assays and in immunohistological sections
*The antibody has applications in studies of cell-cell interactions
*The antibody inhibits cellular adhesion
*Western blotting
*Studies of CD106 in paraffin embedded formalin fixed tissues following high temperature unmasking
PRESENTATION The monoclonal is presented at a concentration of 200µg/2ml in phosphate buffered saline containing 10mM sodium azide and 1mg/ml bovine serum albumin. We recommend that each laboratory determine an optimum working titre for use in its particular application.
STORAGE For use within 1 month of purchase store at +4oC, for long term storage aliquot antibody into small volumes and store at -20oC.
