Description: Monoclonal anti-calbindin D-28k is a mouse IgG produced by hybridization of mouse myeloma cells with spleen cells from mice immunized with calbindin D-28k purified from chicken gut. The antibody was evaluated for specificity and potency:a) by indirect immunofluorescent or immunoperoxidase labelling as well as PAP and Biotin-Avidin labeling of cryostat or vibratome-sections of 4% paraformaldehyde fixed tissue; b) by immunoenzymatic labeling of immunoblots; c) by radioimmunoassay.

The product is a monoclonal antibody (McAB) against calbindin D-28k, and calcium-binding protein of the EF- hand family related to calmodulin and troponin-C. The antibody reacts specifically with calbindin D-28k in tissue originating from human, monkey, rabbit, rat, mouse and chicken (but probably not fish). This antibody does not cross-react with calretinin or other known calcium binding-proteins. This McAB specifically stains the 45Ca-binding spot of calbindin D-28k (MW 28'000. IEP 4.8) in a two-dimensional gel. In radioimmunoassay it detects calbindin D-28k with a sensitivity of 10 ng/assay and an affinity of 1.6 x 10(12) L/M.

Dilution: -Immunohistochemistry: 1:5000 - 1:10,000 with avidin- biotin methods.

-Immunoblots: 1:100 - 1:1,000.

We recommend that the optimal dilutions be determined by titration experiments.

Storage: For long storage, keep at -80'C (or at least -20'C). For continuous use keep at 4'C (with 0.01% Na-azide). Avoid repeated freezing and thawing.

Description: This antiserum was produced against recombinant rat calbindin D-28k. It cross reacts with many other species, including humans. It can be used for immunohistochemistry and immunoblotting. In immunoblots it has 10% crossreaction with calretinin. Thus, depending on the dilution of the antiserum and/or sensitivity of the immunostaining, this other calcium- binding protein could be also revealed. In case of doubt, we suggest to use our monoclonal antibody cat#RDI-CALBINDabm directed against calbindin D-28k, which does not recognize calretinin.

dilutions: Immunohistochemistry: 1:10,000 - 1:20,000 with the avidin-biotin methods.

Immunoblots: 1:100 - 1: 1,000

Storage: Reconstitute by shaking very long and sucking with a Pasteur-pipette until the pellet is dissolved. Make small portions (e.g.2-5 ul). For long storage, keep at -80'C (or at least -20'C). For continuous use keep at 4'C (with 0.01% Na-azide). Avoid repeated freezing and thawing.
