Activity: Reacts with basigin or neurothelin, a transmembrane glycoprotein (50-60kd) of the immunoglobulin super-gene family. Neurothelin is a blood brain barrier-specific molecule. It bears homology with mouse gp42 or basigin, human M6 or EmMprin, rat OX-47 or CE-9, and avian HT7 or 5A11. Neurothelin is a molecule which is broadly expressed on cells of hematopoietic and non-hematopoietic origin. Its expression on specific cell types may be regulated by cytokines. CD147 plays a role in embryonal blood-brain barrier development and a role in integrin-mediated adhesion in brain endothelia. Binds to epitope 1 of CD147.
Application: -Indirect immunofluorescence cell surface staining (flow cytometry) , Titer for each application
(does not inhibit homotypic aaggregation or adhesion).

SPECIFICITY The antibody recognizes the 50-60kDa CD147 antigen that is broadly expressed on cells of both haematopoietic and non-haematopoietic derivation. The antigen is upregulated upon cell activation in particular on lymphocytes and monocytes.
APPLICATIONS * Flow cytometry* Studies on tumour cells where CD147 is believed to bind a fibroblast ligand which stimulates the production of collagenase and other extracellular matrix metalloproteinases.

SPECIFICITY: This antibody recognizes 50-60Kda cell surface protein which is the human homologue of rat OX-47 antigen. (This moiety is expressed in blood brain barrier endothelium, on normal and neoplastic breast tissue)
APPLICATIONS: *Flow Cytometric studies of the expression of CD147 (also known as neurothelin)
PRESENTATION: Each monoclonal is delivered in a sealed vial at a concentration of not less than 200µg/2ml in in phosphate buffred saline containing 10mM sodium azide and 1mg/ml bovine serum albumin. We recommend that each laboratory determine an optimum working titre for use in its particular application.
STORAGE: Purified monoclonal antibodies are generally stable for long periods at 4oC. However repeated warming to room temperature and re-cooling may result in a loss of activity and hence effective working titre. It is recommended that monoclonal antibodies are aliquotted upon receipt and then stored at -20C. After thawing they should then be used as a stock solution for up to 60 days. Dilute working solutions are best prepared on the day of use.

Application: 500K cultured Raji cells per tube were washed and incubated 45 minutes on ice with 80ul of the anti- CD147:PE antibody diluted 1:50 at a concentration of 10ug/ml. Cells were washed three times, fixed and analyzed using a BD FACSCalibur. Cells stained positive with a mean shift of 2.26 log 10 fluorescent units when compared to a mouse IgG1:PE
