Background: The NA+/K+ Atpase is an integral membrane enzyme found in all cells of higher organisms and is responsible for the ATP-dependent transport of NA and K across the cell membrane. The enzyme consists of a large multipass transmembrane catalytic subunit (alpha subunit) and an associated smaller glycoprotein (Beta subunit). Studies indicate that there are three isoforms of the alpha subunit (A1, a2, & A3) and two isoforms of the Beta subunit (B1 & B2) which are encoded by two multigene families. The different isoforms of the NA/K ATPase exhibit tissue specific and developmental pattern of expression. The Alpha 1 and Beta mRNAs are present in all cell types examined whereas the Alpha-2 and Alpha-3 mRNAs exhibit a more restricted pattern of cell-specific expression.

Storage: Store at -20 DEG C upon receipt. Avoid frequent freeze thaw cycles. Recommend aliquoting. BEFORE OPENING, suggest centrifuging for 1 minute at 1000 RPM to concentrate material in vial.

Packaging: 200ug protein G purified mouse IgG2a 0.1M Tris-glycine pH 7.4 with 0.05% sodium azide). -After thawing, tighten cap and centrifuge for 1-2 minutes at 1000 RPM to concentrate antibody in vial. Further dilutions in Tris-glycine buffers or PBS with 0.05% NaN3 (containing 1% BSA or carrier protein recommended).

Uses: Western blot: 0.05-1ug/ml dilution in blocking solution (TRis buffered saline containing 3% non fat dry milk . Detects differentially glycosylated isoforms of Na-K ATPase Beta 1 isoform at approx 55/60 kda in brain, heart and kidney microsomal preparations
