RDI Division of Fitzgerald Industries Intl  offers a wide line of  antibodies. Since no one antibody works best for all applications (neutralization, blotting, ELISA, etc), we offer many different types of antibodies to help solve this problem. mouse monoclonal anti-human 29 kD ANCA- antigen

Specificity:  Ag. MW.: 29 kD.

Reactivity: The monoclonal antibody reacts with the human myeloid lysosomal serine protease, also known as '29kD c-ANCA-antigen', but not with other serine proteases such as elastase and cathepsin G (1).

The monoclonal reacts with human cells of the myeloid lineage, from early promyelocytes to neutrophils and monocytes. The monoclonal antibody does react with neutrophils and monocytes from chimpanzees, but not with cells from macacus fascicularis and other species less closely related to man.

Applications: Antigen catching ELISA for specific detection of human 29kD-ANCA (c-ANCA) (1).

Methods: Indirect immunofluorescence staining with analysis by FACS or fluorescence microscopy (2). Immunohistology on cryopreserved- or formalin-fixed tissue (3). Immunoelectron microscopy using cryopreserved or 4% PFA, 0.5% glutaraldehyde fixed cells (4).

Clone: CLB-702.

This clone has been derived from hybridization of SP2/0 cells with spleen cells of a BALB/c mouse immunized with extract from neutrophil azurophilic granules.

Mouse isotype:The monoclonal antibody is of the mouse subclass IgG1.

Source: Ascites fluid of tumor bearing BALB/c mice.

Purification: Ammoniumsulphate precipitation and ion exchange chromatography.

Packing: Each vial contains 1 ml with approximately 0.2mg/ml monoclonal antibody and 10 mg BSA in 20 mM TRIS and 150 mM NaCl, pH 8.0.

Preservative: Merthiolate (0.001%).


Storage: Monoclonal antibodies should be stored below -18Deg C. Repeated thawing and freezing may reduce the antibody activity and should be avoided. Do not use this product beyond the stated expiration date.

This clone has bee derived from hybridization of Sp2/0 myeloma cells with spleen cells of a Balb/c mice immunized with human proteinase PR3 purified leukocyte mass

Application: Antibodies can be used in ELISA

Purification: Purified by chromatogrpahy on protein A Sepharose. Purity is tested by electrophoresis.

Packaging: 1mg in ml (mg/ml) PBS pH7.4 with 0.1% sodium azide.

NOTE: Recommend centrifuging the unopened vial at 500-1000 RPM for 2 minutes to concentrate antibody in bottom of vial. If desired, you can add PBS containing 0.1% NaN3 to bring volume to 1mg in 1ml.

DESCRIPTION. Highly purified by chromatography. Free of   elastase and cathepsin G. Prepared from 1mM Citric acid pH 3.5 + 1 M NaCl and 0.005% Triton® X100 ,lyophilization after dialysis vs. 1mM Citric acid pH 3.5 + 0.005% Triton(tm) X 100. The dried product contains approximately 10% PR3 protein and 90% solids (sold on basis of PR3 content).   Re-dissolve with H2O to 1 mg/ml.

SOURCE. Leucocytes of Purulent Human Sputum.

MW: SDS-PAGE indicates MW = 29,000.

Purity > 95% by native electrophoresis at pH 4.3 (4) yielding predominately 2 bands that migrate one-half the distance in comparison to Cathepsin G. Elastase, Lysozyme and Myeloperoxidase are also compared. A similar electrophoretic pattern was first described by Baggiolini et.al. (2) for the three proteases.

Storage: Store frozen at -20°. Re-dissolve with H2O to 1 mg/ml. After reconstitution,may be freeze-thawed up to 5 times (avoid frequent freeze thaw cycles, recommend aliquoting and store -20 DEG C).

Activity Test: 35 - 45 units per mg of protein. One unit will hydrolyze 1mMole of Boc-Ala-Ala-Nva-SBzl per minute at pH 7.5 at 25°C. The assay buffer-substrate (2 ml) is 0.1 M MOPS pH 7.5 containing 0.5 M NaCl, 10% DMSO, 1 mM substrate and 0.1 mM 5,5'-dithiobis-(2-nitrobenzoicacid). Use 0.2 to 0.5 ug of enzyme. Read increase in absorbance at 405 nm. The 1.0 mM extinction coefficient is 13.1 in a 10 mM cell.The kinetics of PR3 hydrolysis of thiolbenzyl substrates is described by Kam et. al. (1)
