Since no one antibody works best for all applications (neutralization, blotting, ELISA, etc), we offer many different types of antibodies to help solve this problem. Presentation: 100ug antigen affinity purified IgG (lyophilized) from 1mg/m PBS 7.4

Immunogen: recombinant human BMP-2

Purification: affinity chromatography using immobilized rec human BMP-2 matrix

Reconstitution:add 0.5ml sterile DI water to yield a 200ug/ml stock.

Storage: Stable at least 4 weeks at 2-4 DEG C under aseptic conditions. If not used for cell/neutralization then recommend adding 0.05-0.09% sodium azide or other suitable preservative. Further dilution in PBS with 0.1% BSA or other carrier recommended. -Long term: aliquot and store -20 DEG C. Avoid frequent freeze thaw cycles

Uses: -Western blot: approx 0.2-0.5ug/ml (load 50ug whole cell lysate or 100-10ng/lane of pure rec human BMP-2 with chemiluminescence detection

-Elisa:

-Neutralization: ND50 approx 2ug/ml with use of 200ng/ml BMP-2

-Histochemistry: not tested, would recommend titration from 1-10ug/ml on acetone fixed frozen sections.

Goat anti-BMP-2/4 (Human/Rat)

Presentation: affinity purified goat IgG at 1mg/ml in PBS   with 5% Trehalose

Immunogen: recombinant human BMP-2

Reactivity: human and rat (others not tested)

Use: -western blot: 0.1-0.2ug/ml with chemiluminescence detection)

         -Histochemistry: approx 5-15ug/ml

        -Elisa:0.5-1ug/ml

Optimal dilutions should be determined by user for each application

Storage: Store frozen. Aliquot as undiluted antisera and immediately place at -20 DEG C. At first thaw, tighten cap, centrifuge for 1-2 minutes at 1000 rpm to concentrate material in vial. Stable at least 6 months at -20 DEG C. Avoid repeated freeze/thaw cycles.

Specificity: This antibody has been selected for its ability to recognize BMP-2 and BMP-4 in direct ELISA and western blot assays.

-Immunocytochemistry: This antibody can be used with the appropriate secondary reagents to detect human BMP-2/BMP-4. An experimental protocol is listed below. Cells (cultured or recently collected) may be fixed for 20 minutes at room temperature with freshly prepared 1 - 2% paraformaldehyde/PBS (pH 7.4). Three to five washes of cells in PBS (15 minutes each) is usually required after fixation and before application of primary antibodies. Labeling may be obtained by incubating cells overnight at 4 ° C with 2 -5 g/mL anti -human BMP-2/4 antibodies. Immunohistochemistry: Tissues may be dissected from experimental animals that were fixed by vascular perfusion with 4% paraformaldehyde/PBS (pH 7.4) and followed by perfusion with a 10% sucrose solution in 0.1 M phosphate buffer (pH 7.2). Adequate labeling may be achieved on 5 -15 m thick cryostat sections by incubating them with primary antibodies diluted to 5 -15 g/mL.On free -floating sections, primary antibodies should be diluted to 0.5 -reduce background staining.

Western Blotting: This antibody can be used at 0.1 -0.2 g/mL with the appropriate secondary reagents to detect human BMP -2 and human BMP-4. The detection limit for recombinant human BMP-2 and recombinant human BMP-4 is approximately   200 ng/lane under non reducing and reducing conditions.

Mouse anti-Human Bone Morphogenetic Protein (BMP)

Mouse anti-Bone Morphogenetic Protein 4 (BMP-4) (monoclonal)

Isotype: mIgG2b

Immunogen: Recombinant mouse bone morphogenetic protein 4.

Specificity: Specific for bone morphogenic protein-4 (BMP-4) by Western Blot and Elisa. No cross reactivity with human BMP-2 or human TGF-B1. The antibody reacts with both mouse BMP-4 in both reduced and non-reduced conditions.

Applications: Immunohistochemistry demonstrated the presence of BMP-4 in developmental bone formation in human embryo growth plate and alveolar bone of rat embryo. Immunocytochemistry using osteoinductive cells, i.e., BFP and Saos-2 Western Blot using osteoinductive cells; i.e., BFP and Saos-2 Optimal working dilutions must be determined by the end user.

Storage: Store at 2-8 oC for up to 6 months from date of receipt

Application:

BMP4 is one of nine structurally related BMPs belonging to the transforming growth factor beta (TGF-Beta) superfmaily of secreted proteins. Mature BMP4 is a dimer that binds to a multimeric transmembrane receptor with serine/threonine kinase activity. although BMP4 was discovered because it stimulates bone formation in adult mammals, it has important roles as signaling molecule in embroyonic tissues including the developing central and peripheral nervous system, musculature and skeleton. It also participates in a signaling pathway fund in insects and worms. BMP-4 is implicated in fibrodysplasia osificans progressiva. It is uniquely overexpressed in lymphoblastoid cells and preosseous fibroproliferative lesions. BMPs 1 to 6 are expressed in prostatic adenocarcinoma and BMPs 1 to 5 are expressed in both benign prostatic hyperplasia and occular melanoma. BMP4 also plays an important role in early tooth morphogenesis.
