Immunogen: A 17-amino acid synthetic peptide from the carboxyl-terminus of RAT TGF .

Hybridoma: Produced by fusion between BALB/c splenocytes and myeloma p3X63/Ag8.653 cells.

Use: Western Blotting (Non-reduced only)

         Immunohistology (Frozen & formalin/paraffin)

Background: It recognizes a 6kDa polypeptide, identified as the transforming growth factor (TGF ).1 Ab-1 shows no cross-reaction with EGF and the neuropeptide synenkephalin.4 TGF (aa 50) is a growth factor with 33% homology to EGF, binds to EGFR, activates tyrosine phosphorylation of the receptor, and stimulates cell proliferation. It plays a role in tumor initiation by inducing the reversible transformed phenotype. Overexpression of TGF-alpha may be important for tumor progression via autocrine stimulation and oncogene overexpression. TGF bioactivity is reported in the urine of cancer   patients.

Preparation :  Produced from sera of rabbits pre-immunized with highly pure (>98%) recombinant hTGF-alpha. Anti-hTGF-alpha specific antibody was purified by affinity chromatography employing immobilized hTGF-alpha matrix.

Formulation :  A sterile filtered antibody solution (1mg/ml) was lyophilized from 0.5X PBS pH 7.4.

Reconstitution :  The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -200C. When reconstituted in sterile water to a concentration of >0.2 mg/ml the antibody is stable for at least six weeks at 2-40C.

Applications :

Neutralization -To yield one-half maximal inhibition [ND50] of the biological activity of hTGF-alpha (2.0 ng/ml), a concentration of 0.10 - 0.15 µg/ml of this antibody is required.

Elisa -To detect hTGF-alpha by direct ELISA (using 100µl/well antibody solution) a concentration of at least 0.5µg/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with compatible secondary reagents, allows the detection of 0.2 - 0.4 ng/well of recombinant hTGF-alpha.

Western Blot  To detect hTGF-alpha by Western Blot analysis this antibody can be used at a concentration of 0.1 - 0.2 µg/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hTGF-alpha is 1.5 - 3.0 ng/lane, under either reducing or non-reducing conditions.
