Background: Superoxide dismutase (SOD) is responsible for the elimination of cytotoxic active oxygen by catalyzing the dismutation of the superoxide radical to oxygen and hydrogen peroxide. There are two types of SOD in mammalian cells. One form contains Cu and Zn ions and exists in the cytosol.

Size: 1.0mg IgG fraction. 1 mg/ml (1ml volume). Liquid frozen. Formulated in 0.025 M NaC1/0.1 M Tris-HCl, pH 7.5.

Purity: Anti-Superoxide Dismutase (Cu/Zn-SOD) IgG fraction is a polyclonal antibody raised in goat against purified recombinant yeast-derived SOD, and purified by sequential chromatography, using specific precipitation and ionic exchange chromatography.

Biological Activity: This polyclonal antibody recognizes human SOD in a western analysis (1/100 dilution) and ELISA.

Storage: 1 year at -20'C.

RECOMBINANT HUMAN SUPEROXIDE DISMUTASE (SOD)

Formulation: 10 mg/ml in water (no additive is added).

Description: Cu/Zn Human Superoxide Dismutase is a stable dimer of identical subunits with a combined molecular mass of 32,000 daltons. This enzyme dismutes the superoxide radical to molecular oxygen. This enzyme has been expressed in yeast and purified using sequential chromatography steps. The hSOD is acetylated at its N-terminus.

Source: Human SOD is produced in genetically engineered yeast, and purified by ionic exchange chromatography.

MW: 32 kDa (dimeric protein)

Biological Activity: The specific activity of SOD is 260 U/mg. The enzymatic activity of human SOD is determined by calorimetrically determining the oxidation of a chromophore. This was performed with a commercially available kit called SOD-525 from Biooxytech S.A. (Cedex,France).

Purity: The material has been judged to be >98% pure, as determined by non-reduced SDS gel electrophoresis.

Stability: Human recombinant SOD is stable at -20'C for several months.

Storage: Upon receipt, the material should be stored at -20'C.

Sheep Anti-Superoxide Dismutase (Mn-SOD)

Background: Superoxide dismutases are found in all aerobic organisms. Their physiological function appears to be to protect cells from free radicals by scavenging superoxide anions. They catalyse the dismutation of superoxide anions to oxygen and hydrogen peroxide. SOD (Mn) is located primarily in the mitochrondria and is thought to be of   prokaryote origin. SOD(Mn) is a tetramer of   identical subunits of MW 25,000 by SDS-PAGE.


Size: 1ml Sheep IgG fraction in glycine buffered saline pH 7.4 with 0.1% NaN3 and 1mM EDTA

Antigen: human superoxide dismutase Mn enzyme purified from human liver.


Purification: ion exchange chromatography


Specificity: reacts specifically with human MnSOD. Monospecificity is performed by testing the antiserum by immunoelectrophoresis, two dimensional IEP and double diffusion.


Activity: -Useful in double diffusion: 10ul antiserum vs 10ul SOD(Mn). The use of 3% PEG 6000 with 1.2% agarose in a suitable buffer (as TBE or Tris- barbital pH >8.2).
            --Western blot:approx 1:200 dilution against a 10% liver cell lysate

              -histochemistry

Storage: 2-8 DEG C. Slight precipitation can occur on storage which may be removed by centrifugation and should not affect performance characteristics.
