DATA SHEET:recombinant Human B-NGF

Beta-NGF is a neurotrophic factor structurally related to BDNF, NT-3 and NT-4. These proteins belong to the cysteine-knot family of growth factors that assume stable dimeric structures. Beta-NGF is a potent neurotrophic factor that signals through its receptor beta-NGFR, and plays a crucial role in the development and preservation of the sensory and sympathetic nervous systems. Beta-NGF also acts as a growth and differentiation factor for B lymphocytes and enhances B-cell survival. The functional form of human beta-NGF is a noncovalently disulfide-linked homodimer, of two 13.5 kDa polypeptide monomers (238 total amino acid residues). The three disulfide bonds are required for biological activity

Reconstitution: The lyophilized human B-NGF is soluble in water and most aqueous buffers. The material can be reconstituted to a concentration of 0.1-1mg/ml in water. Allow to set at least 30 minutes at 4 DEG C. Tighten cap, mix gently by inversion. If desired, can centrifuge for 1-2 minutes at 1000RPM to concentrate material in vial. This solution can be diluted into water or other buffered solutions or stored at 4 DEG C for up to 1 week or at -20 DEG C for future use.

Note:If this material is not to be used for coating or labeling (or other applications requiring carrier free material), the addition of 0.1% BSA to dilution buffers or dilution in sterile culture media is recommended to enhance stability and minimize absorption to vial. Ideally, maintain stock >10ug/ml.

Storage: The lyophilized powder, though stable at room temperature for a few weeks, is best stored dessicated at -20 DEG C. Reconstituted B-NGF should be stored in working aliquots at -20 DEG C. Avoid Frequent Freeze- THAW Cycles.

Reactivity: Assay#1: The Ed50 as determined by the dose-dependent stimulation of the   proliferation of human TF-1 cells is <1ng/ml, cprresponidng to a specific   activity of 1million units/mg continuned on back side

Assay#2: Determine dby its ability to stimulate chick E9 DRG neurite outgrowth. The ED50 is <1ng/ml corresponding to a specific activity of >1million units/mg.
