DATA SHEET:  recombinant Human Fas(receptor)

Fas and Fas Ligand (FasL) belong to the TNF superfamily and are type I and type II transmembrane proteins, respectively. The Fas molecule is comprised of 319 amino acids of which 145 amino acids constitute the cytoplasmic region, 17 amino acids the transmembrane domain and 157 amino acids the extracellular region. The extracellular portion of Fas is called sFasR. Binding of FasL to Fas triggers apoptosis in Fas-bearing cells. The mechanism of apoptosis involves recruitment of pro-caspase 8 through an adaptor molecule called FADD and processing of the pro-enzyme to active forms. These active caspases then cleave various cellular substrates leading to the eventual cell death. sFasR is capable of inhibiting FasL-induced apoptosis by acting as a decoy receptor that serves as a sink for FasL. Recombinant Human sFasR is a 17.6 kDa protein consisting of 157 amino acid residues.

Reconstitution: The lyophilized sFASR is soluble in water and most aqueous buffers. The lyophilized powder should be reconstituted in sterile DI water, to a concentration of 0.1-1mg/ml. Allow to set at least 30 minutes at 4 DEG C. If desired, can centrifuge for 1 minute at 1000 rpm to concentrate material in vial. This solution can be diluted into water or other buffered solutions or stored at 4 DEG C for up to 1 week or at -20 DEG C for future use. For most in-vitro applications,

Note:If this material is not to be used for coating or labeling (or other applications requiring carrier free material), the addition of 0.1% BSA to further dilution buffers or dilution in sterile culture media is recommended to enhance stability and minimize absorption to vial. Ideally, maintain stock >10ug/ml.

Storage: The lyophilized powder, though stable at room temperature for a few weeks, is best stored dessicated at -20 DEG C. Reconstituted sFas-R should be stored in working aliquots at -20 DEG C. Avoid Frequent Freeze-THAW Cycles.

Reactivity: The Ed50 was determined by its abaility to inhibit the cytotoxicity of Jurkat cells is between 10-15ug/ml in the presence of 2ng/ml of hFASl.
