DATA SHEET:recombinant Human MIP-1 Alpha (Macrophage Inflammatory Protein 1 alpha)

Both MIP-1 alpha and MIP-1 beta are structurally and functionally related CC chemokines. They participate in the host response to invading bacterial, viral, parasite and fungal pathogens by regulating the trafficking and activation state of selected subgroups of inflammatory cells e.g. macrophages, lymphocytes and NK cells. While both MIP-1 alpha and MIP-1 beta exert similar effects on monocytes their effect on lymphocytes differ; with MIP-1 alpha selectively attracting CD8+ lymphocytes and MIP-1 beta selectively attracting CD4+ lymphocytes. Additionally, MIP-1 alpha and MIP-1 beta have also been shown to be potent chemoattractants for B cells, eosinophils and dendritic cells. Both human and murine MIP-1 alpha and MIP-1 beta are active on human and murine hematopoietic cells. This recombinant human MIP-1 alpha is a 7.8 kDa protein containing 69 amino acid residues, including the four highly conserved cysteine residues present in CC chemokines.

Purity: >99% by SDS-Page analysis and RP-HPLC analysis. Endotoxin level is less than 0.1ng per ug of MIP-1 alpha.

Reconstitution: The lyophilized MIP-1A is soluble in water and most aqueous buffers. The lyophilized powder should be reconstituted in water to a concentration of 0.1 milligram per ml. Allow to set at least 30 minutes at 4 DEG C , tighten cap and rotate to reconstitute any material on sides of vial. If desired, you can centrifuge at 1000 rpm for 1 minute to concentrate volume in bottom of vial. This solution can be diluted into water or other buffered solutions or stored at -4 DEG C for up to 1 week or at -20 DEG C for future use.

Note: If this material is not to be used labeling(or other applications requiring carrier free material), the addition of 0.1% protease free BSA to dilution buffers or dilution in sterile culture media is recommended to enhance stability and minimize absorption to vial.

Storage: The lyophilized powder, though stable at room temperature, is best stored desiccated below 0 DEG C. reconstituted MIP-1alpha should be stored inw orking aliquots at -20 DEG C. Avoid Frequent Freeze-THAW Cycles.

Reactivity: Determined by it ability to chemoattract human monocytes using a  concentration range of 1.0-10.0ng/ml.
