DATA SHEET:recombinant Human MCAF (MCP-1)

-Human macrophage/monocyte chemotactic and activating factor (MCAF) also called human macrophage chemoattractant protein-1 (MCP-1) is an 8.6 kDa protein containing 76 amino acid residues. It plays an important role in the inflammatory response of blood monocytes and tissue macrophages.

Purity: >98% by SDS-Page analysis and HPLC analysis. Endotoxin level is <0.1ng per ug (1EU/ug) of MCAP.

Reconstitution: The lyophilized MCAF/MCP-1 should be reconstituted in water to a concentration of 0.1-1mg/ml. Allow to set at least 30 minutes at 4 DEG C, Mix well. If desired, centrifuge for 1-2 minutes ta 1000rpm to concentrate material in vial. This solution can be diluted into other buffered solutions or stored at 4 DEG C for up to 1 week or at -20 DEG C or future use. For most in-vitro applications, MCP-1 exerts its biological activity in the range of 10.0 to 100 ng/ml.

Note:If this material is not to be used for coating or labeling (or other applications requiring carrier free material), the addition of 0.1% protease free BSA to dilution buffers or dilution in sterile culture media is recommended to enhance stability and minimize absorption to vial. Ideally, maintain stock >50ug/ml.


Storage: The lyophilized powder is stable at room temperature for a few weeks but it is best stored desiccated at -20 Deg C. Reconstituted MCAF should be stored in working aliquots at -20DEG C. AVOID FREQUENT FREEZE THAW CYCLES.

Reactivity: Determined by its ability to chemoattract human monocytes using a concentration range of 10- 100ng/ml.

Appearance: Sterile Filtered White lyophilized (freeze-dried) powder. Lyophilized from a concentrated (1mg/ml) solution in water containing no additives.

Purity: Greater than 99.0% as determined by:

(a) Analysis by RP-HPLC.

(b) Anion-exchange FPLC.

(c) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel.

Dimers and aggregates:

Less than 1% as determined by silver-stained SDS-PAGE gel analysis.

Endotoxin: Less than 0.1 ng/µg (IEU/µg) of rHuMCP-1.

Solubility: It is recommended to reconstitute the lyophilized rHuMCP-1 in sterile 18MO-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.

Stability: Lyophilized rHuMCP-1 although stable at room temperature for 3 weeks, should be stored desiccated below -18 C. Upon reconstitution rHuMCP-1 should be stored at 4 C between 2-7 days and for future use below -18 C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please avoid freeze-thaw cycles.

Sequence: The sequence of the first five N-terminal amino acids was determined and was found to be Gln-Pro-Asp-Ala-Ile.

Biologicval Activity MCP-1 is fully biologically active when compared to standard. The specific activity as determined by the ability of MCP-1 to chemoattract human Monocytes using a concentration of 5-20 ng/ml.

Protein content: Protein quantitation was carried out by two independent methods:

1. UV spectroscopy at 280 nm .

2. Analysis by RP-HPLC, using a standard solution of MCP-1 as a Reference Standard.
