DATA SHEET:recombinant Human GRANULOCYTE MACROPHAGE COLONY STIMULATING FACTOR

GM-CSF is a hematopoietic growth factor that stimulates the development of neutrophils and macrophages and promotes the proliferation and development of early erythroid megakaryocytic and eosinophilic progenitor cells. It is produced in by endothelial cells, monocytes, fibroblasts and T-lymphocytes. GM-CSF inhibits neutrophil migration and enhances the functional activity of the mature end-cells. The human and murine molecules are species-specific and exhibit no cross-species reactivity. This recombinant human GM-CSF is a 14.6 kDa globular protein consisting of 128 amino acids containing tow intramolecular disulfide bonds and two potential N-linked glycosylation sites..

Reconstitution: The lyophilized GM-CSF is soluble in water and most aqueous buffers. The lyophilized powder should be reconstituted in water or 10mM sodium citrate pH3.5 to a concentration of 0.1-1.0 mg/ml. Allow to set at least 30 minutes at 4 Deg C, rotate, mix well. If desired, centrifuge for 1-2 minutes at 500- 1000 rpm to concentrate material in vial.This solution can be diluted into other buffered solutions or stored at 4 DEG C for up to 1 week or at -20 DEG C for future use. For most in-vitro applications, human GM-CSF exerts its biological activity in the concentration range of 0.05 to 0.5 ng/ml.

Note: For material that will not be used for coating or labelling (or other applications requiring carrier free material), we recommend adding 0.1% protease free BSA/HSA or media to any dilution buffers. Maintain stock >10ug/ml.

Storage: The lyophilized powder, though stable at room temperature for a few weeks, is best stored dessicated at -20 DEG C. Reconstituted GM-CSF should be stored in working aliquots at -20 DEG C. Avoid Frequent Freeze-THAW Cycles.

Reactivity: The Ed50 as determined by the dose dependent stimulation of the proliferation of human TF-1 cells is <0.1ng/ml, which corresponds to a specific activity of > 1 X 10 E07 units/mg.
