-TNF-beta is a potent mediator of inflammatory and immune responses. It belongs to the TNF family of ligands and signals through TNFR1 and TNFR2. TNF-beta is produced by activated T and B lymphocytes, and has similar activities to TNF-alpha. Like TNF-alpha, TNF-beta is involved in the regulation of various biological processes including cell proliferation, differentiation, apoptosis, lipid metabolism, coagulation, and neurotransmission. TNF-beta is secreted as a soluble polypeptide, but can form heterotrimers with lymphotoxin-beta, which effectively anchors the TNF-beta to the cell surface. TNF-beta is cytotoxic to a wide range of tumor cells. This recombinant human TNF-beta is a secreted 171 amino acid protein (18.6 kDa) which forms an active non-disulfide linked homotrimer structure in solution

Reconstitution: The lyophilized TNF-B should be reconstituted in sterile water to a concentration of 0.1-1mg/ml. Allow to set at least 30 minutes at 4 DEG C, tighten cap and mix well. If desired, centrifuge 1 minute at 1000 rpm to concentrate materialin vial. This solution can be diluted into other buffered solutions an dstored at 4 DEG C forup to 1 week or stored at -20 DEG C for future use. For most in -vitro applications, TNF-B exerts its biological activity in the range of 0.05 to 20.0 ng/ml.

Note: If this material is not to be used for coating or labeling (or other applications requiring carrier free material), the addition of 0.1% BSA to dilution buffers or dilution in sterile culture media is recommended to enhance stability and minimize absorption to vial. Ideally, maintain stock >50ug/ml.

Storage: The lyophilized powder is stable at room temperature for a few weeks but it is best stored desiccated at -20 DEG C.. Reconstituted TNF-B should be stored in working aliquots at -20DEG C. AVOID FREQUENT FREEZE THAW CYCLES.

Reactivity: The Ed50 as determined by the cytolysis of murine L929 cells in the presence of Actinomycin D is <0.05ng/ml, correspponding to a specific activity of > 2 X 10 E07 units/mg.
