DATA SHEET:recombinant Human INTERLEUKIN-6

-IL-6 is a pleiotropic cytokine that plays an important role in host defense by regulating immune and inflammatory responses. Produced by T cells, monocytes, fibroblasts, endothelial cells and keratinocytes, IL-6 has diverse biological functions. It stimulates B-cell differentiation and antibody production, synergizes with IL-3 in megakaryocyte development and platelet production, induces expression of hepatic acute-phase proteins, and regulates bone metabolism. IL-6 signals through the IL-6 receptor system that consists of two chains, IL-6R alpha and gp130. Murine IL-6 is inactive on human cells, while both human and murine are equally active on murine cells. Recombinant human IL-6 is a 20.9 kDa protein containing 184 amino acid residues.

Supplied: Lyophilized powder with no additives. Endotoxin level is less than 0.1ng per ug of IL-6.

Purity: >98% by SDS-Page analysis and HPLC analyses. Endotoxin level is less than 0.1ng per ug (1EU/UG) of IL-6.

Reconstitution: The lyophilized IL-6 dissolves very slowly in water or buffered solutions at physiological pH. To accelerate this solution of lyophilized IL-6 use the following protocol: a) Suspend the lyophilzied material in water to a concentration of 0.1- 0.5mg/ml.

b) Add acetic acid to a concentration of 15mM.

c) Allow to set at least 5 minutes. Rotate and/or vortex to mix.

Centrifuge for 1-2 minutes at 1000 rpm to concentrate solution in bottom of vial. This solution can be diluted into other buffered solutions or stored at 4 DEG C for up to 1 week or at -20 DEG C for future use.

Note::If this material is not to be used for coating or labeling or other applications requiring carrier free material), the addition of 0.1% BSA to dilution buffers or dilution in sterile culture media is recommended to enhance stability and minimize absorption to vial.

Storage: The lyophilized powder is stable at room temperature for a few weeks but it is best stored desiccated at -20 Deg C. Reconstituted IL-6 should be stored in working aliquots at -20 DEG C. AVOID FREQUENT FREEZE THAW CYCLES.

Reactivity: The ED50 as determined by the dose-dependent stimulation of the proliferation of IL-6 dependent murine 7TD1 cells is <0.1ng/ml, corresponding to a specific activity of > 1 X 10 E07 units/mg.

Description : Recombinant Human IL-6 produced in E.Coli is a single, non-glycosylated polypeptide chain containing 184 amino acids and having a molecular mass of 21000 Dalton. The rHuIL-6 is purified by proprietary chromatographic techniques.

Appearance: Sterile Filtered White lyophilized (freeze-dried) powder

Endotoxin: Less than 0.1 ng/µg (IEU/µg) of rHuIL-6.

Formulation: Lyophilized from a concentrated (1mg/ml) solution in water containing no additives.

Purity: Greater than 98.0% as determined by:

(a) Analysis by RP-HPLC.

(b) Anion-exchange FPLC.

(c) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel.

Amino acid sequence: The sequence of the first five N-terminal amino acids was determined and was found to be Met-Pro-Val-Pro-Pro.

Dimers and aggregates: Less than 1% as determined by silver-stained SDS-PAGE gel analysis.

Solubility: It is recommended to reconstitute the lyophilized rHuIL-6 in sterile 18MO-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.

Stability: Lyophilized rHuIL-6 although stable at room temperature for 3 weeks, should be stored desiccated below -18 C. Upon reconstitution rHuIL-6 should be stored at 4 C between 2-7 days and for future use below -18 C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please avoid freeze-thaw cycles.

Biological Activity: rHuIL-6 is fully biologically active when compared to standard. The ED50 as determined by the dose-dependant stimulation of murine 7TD1 cells is less then 0.1 ng/ml, corresponding to a Specific Activity 1 X 10 E07 units/mg.

Protein content: Protein quantitation was carried out by two independent methods:

1. UV spectroscopy at 280 nm using the absorbency value of 0.47 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).

2. Analysis by RP-HPLC, using a standard solution of IL-6 as a Reference Standard
