-IL-3 is a hematopoietic growth factor that promotes the survival, differentiation and proliferation of committed progenitor cells of the megakaryocyte, granulocyte-macrophage, erythroid, eosinophil, basophil and mast cell lineages. Produced by T cells, mast cells and eosinophils, IL-3 enhances thrombopoieses, phagocytosis, and antibody-mediated cellular cytotoxicity. Its ability to activate monocytes suggests that IL-3 may have additional immunoregulatory roles. Many of the IL-3 activities depend upon co-stimulation with other cytokines. IL-3 is species-specific, variably glycosylated cytokine. Recombinant human IL-3 is a 15.0 kDa globular protein containing 133 amino acid residues.

Reconstitution: The lyophilized IL-3 should be reconstituted in sterile water to a concentration of 0.1-1.0mg/ml Allow to set at least 30 minutes at 4 DEG C, rotate/mix well. If desired, centrifuge for 1-2 minutes at 1-2K RPM to concentrate material in vial. This solution can be diluted into other buffered solutions or storedat 4 DEG C for up to 1 week or at -20 DEG C for future use. For most in-vitro applications, IL-3 exerts its biological activity in the range of 0.1 to 1.0ng/ml.
Note:Note:If this material is not to be used for coating or labeling (or
other applications requiring carrier free material), the addition of 0.1%
BSA to dilution buffers or dilution in sterile culture media is recommended
to enhance stability and minimize absorption to vial.
Storage: The lyophilized powder is stable at room temperature for several weeks but it is best stored desiccated at -20 DEG C. Reconstituted IL-3 should be stored in working at -20DEG C. AVOID FREQUENT FREEZE THAW CYCLES.
Reactivity: The ED50 as determined by the dose-dependent stimulation of the proliferation of human TF-1 cells is <0.1ng/ml, corresponding to a specific activity of >1 X 10 E 07 units/mg.

Description: Recombinant Human IL-3 produced in E.Coli is a single, non-glycosylated polypeptide chain containing 133 amino acids and having a molecular mass of 15000 Dalton. The rHuIL-3 is purified by proprietary chromatographic techniques.
Appearance: Sterile Filtered White lyophilized (freeze-dried) powder.
Endotoxin: Less than 0.1 ng/µg (IEU/µg) of IL-3
Formulation: The protein was lyophilized from a 1mg/ml solution in water with no additives
Purity: Greater than 98.0% as determined by:
(a) Analysis by RP-HPLC.
(b) Anion-exchange FPLC.
(c) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel.
Amino acid sequence: The sequence of the first five N-terminal amino acids was determined and was found to be Ala-Pro-Ala-Pro-Thr.
Dimers and aggregates: Less than 1% as determined by silver-stained SDS-PAGE gel analysis.
Solubility: It is recommended to reconstitute the lyophilized rHuIL-3 in sterile 18MO-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.
Stability: Lyophilized rHuIL-3 although stable at room temperature for 3 weeks, should be stored desiccated below -18 C. Upon reconstitution rHuIL-3 should be stored at 4 C between 2-7 days and for future use below -18 C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).Please avoid freeze-thaw cycles.
Biological Activity: This rHuIL-3 is fully biologically active when compared to standard. The ED50 as determined by the dose-dependant stimulation of TF-1 cells is less then 0.1 ng/ml, corresponding to a Specific Activity of 1 x 10 E07 IU/mg.
Protein content: Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280 nm using the absorbency value of 0.84 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
2. Analysis by RP-HPLC, using a standard solution of IL-3 as a Reference Standard.
