IL-2 is a powerful immunoregulatory lymphokine produced by T-cells in response to antigenic or mitogenic stimulation. IL-2/IL-2R signaling is required for T-cell proliferation and other fundamental functions which are essential for the immune response. IL-2 stimulates growth and differentiation of B-cells, NK cells, lymphokine activated killer cells, monocytes, macrophages and oligodendrocytes. This recombinant human IL-2 is a 15.5 kDa protein, containing 134 amino acid residues including one intrachain disulfide bond.

Purity: >97% by SDS-Page analysis and HPLC analysis. Endotoxin level is less than 0.1 ng per ug (1EU/ug) of IL-2.
Reconstitution: The lyophilized IL-2 should be reconstituted in 100mM acetic acid to a concentration of 0.1-1.0mg/ml. Allow to set at least 30 minutes at 4 DEG C, Mix Well. If desired, centrifuge for 1-2 minutes at 1000 rpm to concentrate material in vial. This solution can be diluted into other buffered solutions or stored at 4 DEG C for up to 1 week or at -20 DEG C for future use. For most in-vitro applications, IL-2 exerts its biological activity in the range of 0.1 to 1.0 ng/ml.
Note:Note:If this material is not to be used for coating or labeling (or other applications requiring carrier free material), the addition of 0.1% BSA to dilution buffers or dilution in sterile culture media is recommended to enhance stability and minimize absorption to vial. Ideally, maintain stock >50ug/ml.
Storage: The lyophilized powder is stable at room temperature for a few weeks but it is best stored desiccated at -20 DEG C. Reconstituted IL-2 should be stored in working aliquots at -20 DEG C. AVOID FREQUENT FREEZE THAW CYCLES.
Reactivity: The Ed50 as determined by the dose dependent stimulation of murine CTLL-2 cells is <0.1ng/ml. The specific activity is greater than 1 X 107 units/mg as determined by the CTLL assay.

Description: Recombinant Human Il-2 is produced in E.Coli is a single, non-glycosylated mutein (variant form), of human IL-2 polypeptide chain containing 134 amino acids and having a molecular mass of 5517Dalton. This recombinant human IL-2 has a Ser substitute for cysteine at position 126. The rHuIL-2 is purified by proprietary chromatographic techniques.
Appearance: Sterile Filtered White lyophilized (freeze-dried) powder.
Endotoxin: Less than 0.1 ng/µg (IEU/µg) of IL-2.
Formulation: The protein was lyophilized after extensive dialysis against 0.17mg sodium phosphate monobasic and 0.89mg dibasic phosphate buffer pH7.5.
Purity: Greater than 98.0% as determined by:
(a) Analysis by RP-HPLC.
(b) Anion-exchange FPLC.
(c) Analysis by reducing and non-reducing SDS-PAGE

Solubility: It is recommended to reconstitute the lyophilized rHuIl-2 in sterile 20mM acetic acid not less than 100µg/ml, which can then be further diluted to other aqueous solutions.
Stability: Lyophilized rHuIL-2 although stable at room temperature for 3 weeks, should be stored desiccated below -18 C. Upon reconstitution rHuIL-2 should be stored at 4 C between 2-7 days and for future use below -18 C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).Please avoid freeze-thaw cycles.
Biological Activity: IL-2 is fully biologically active when compared to standard. The ED50 as determined by the cytolysis of murine CTLL-2 cells in the presence is <0.065ng/ml, corresponding to a Specific Activity of 16.9 million IU/mg
Protein content: Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280 nm using the absorbency value of 0.614 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
2. Analysis by RP-HPLC, using a standard solution of IL-2 as a Reference Standard.
