DATA SHEET:  recombinant HUMAN Myostatin

Myostatin, also known as Growth Differentiation Factor-8 (GDF-8), belongs to the TGF-beta superfamily. Along with other members of the TGF-beta family, Myostatin plays a crucial role as a regulator for embryonic development and tissue homeostasis in adult animals. Myostatin has the ability to halt muscle growth. Early studies have shown Myostatin to be limited to skeletal muscle; however, it is now known that it is found in different muscles throughout the body including the heart. Since both muscle and adipose tissue develops from the same mesenchymal stem cells, new research has suggested that Myostatin may be involved in reducing and or preventing adiposity and type-2 diabetes. Human Myostatin is a homodimeric protein with each subunit containing 110 amino acid residues with a total molecular weight of 25.0 kDa.

Purity: >98% by SDS-Page analysis. Endotoxin level is less than 0.1ng per ug (1EU/ug) of human Myostatin.

Reconstitution: The lyophilized myostatin is soluble in water and most aqueous buffers. The lyophilized powder should first be quick spun in vial, then reconstituted in water to a concentration of 0.1-1.0mg/ml. Allow to set at least 30 minutes at 4 DEG C. If desired, can centrifuge for 1 minute at 1000 rpm to concentrate material in vial. This solution can be diluted into water or other buffered solutions or stored at 4 DEG C for up to 1 week or at -20 DEG C for future use.

Note:If this material is not to be used for coating or labeling (or other applications requiring carrier free material), the addition of 0.1% BSA to dilution buffers or dilution in sterile culture media is recommended to enhance stability and minimize absorption to vial. Ideally, maintain stock >10ug/ml.

Storage: The lyophilized powder, though stable at room temperature for a few weeks, is best stored dessicated at -20 DEG C. Reconstituted material should be stored in working aliquots at -20 DEG C. Avoid Frequent Freeze-THAW Cycles.

Reactivity: Determined by its abbility to inhibit the   proliferation of MCP-11 cells. The expected ED50 for this effect is 17-25ng/ml
