DATA SHEET:recombinant Human Fibroblast Growth Factor-basic

-FGF-basic is one of 23 known members of the FGF family. Proteins of this family play a central role during prenatal development and postnatal growth and regeneration of a variety of tissues, by promoting cellular proliferation and differentiation. FGF-basic is a non-glycosylated heparin binding growth factor that is expressed in the brain, pituitary, kidney, retina, bone, testis, adrenal gland liver, monocytes, epithelial cells and endothelial cells. FGF-basic signals through FGFR 1b, 1c, 2c, 3c and 4. This recombinant human FGF-basic is a 17.2 kDa protein consisting of 154 amino acid residues.

Purity: >95% by SDS-Page analysis and HPLC analysis. Endotoxin less than 0.1ng per ug (1EU/ug) of FGF-b.

Reconstitution: The lyophilized FGF-b is soluble in water and most aqueous buffers. It is sensitive to acidic conditions. For best stabilty, it should be reconstituted in 5mM Tris 7.6 concentration of 1ug/ul. Allow to set at least 30 minutes, rotate and mix well. Centrifuge for 1-2 minutes at 1000RPM to concentrate solution in bottom of vial. This solution can be diluted into other buffered solutions or stored in working aliquots at 4 DEG C for up to 1 week or in single freeze thaw aliquots at -20 DEG C.. Freeze thaw cycles will result in loss of activity. For most in-vitro applications, FGF-b exerts its biological activity in the range of 0.1 to 10.0 ng/ml.

Note:If this material is not to be used for coating or labeling (or other applications requiring carrier free material), the addition of 0.1% BSA to dilution buffers or dilution in sterile culture media is recommended to enhance stability and minimize absorption to vial. Ideally, maintain stock >50ug/ml.

Storage: The lyophilized powder is stable at room it is best stored desiccated below 0 Deg C. Reconstituted FGF-b should be stored in working aliquots at 4 DEG C or aliquoted to -20 DEG C. . FREEZE THAW CYCLES WILL RESULT In SIGNIFICANT LOSS OF ACTIVITY.

Reactivity: This FGF-b is fully biologically active when compared to standards. The ED50 as determined by the dose-dependent stimulation of thymidine uptake by BaF3 cells expressing FGF receptors is less than 0.5ng/ml, corresponding to a specific activity of > 2 X 10 E06 units/mg..

package: Sterile Filtered White lyophilized (freeze-dried) powder, purified by proprietary chromatographic techniques. The protein was lyophilized from a concentrated (1mg/ml) sterile solution containing 5mM Tris pH=7.5 and 150mM NaCl.

Endotoxin: Less than 0.1 ng/µg (IEU/µg) of FGF-basic .

Purity: Greater than 98.0% as determined by:

(a) Analysis by RP-HPLC.

(b) Anion-exchange FPLC.

(c) Analysis by reducing and non-reducing SDS-PAGE Silver

Stained gel.

Solubility: It is recommended to reconstitute the lyophilized rHuFGF-b in sterile 18MO-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.

Stability: Lyophilized rHuFGF-b although stable at room temperature for 3 weeks, should be stored desiccated below -18 C. Upon reconstitution rHuFGF-b should be stored at 4 C between 2-7 days and for future use below -18 C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please avoid freeze-thaw cycles.

sequence: the sequence of the first five N-terminal amino acids was determined and was found to be Met-Ala-Glu-Gly-Glu.

Dimers and aggregates:   Less than 1% as determined by silver-stained SDS-PAGE gel   analysis.

Activity: rHuFGF-b is fully biologically active when compared to standards. The ED50, calculated by the dose-dependant proliferation of BAF3 cells expressing FGF receptors (measured by 3H-thymidine uptake) is <0.5 ng/ml, corresponding to a specific activity of (2 x10^6) Units/mg.

Protein content:Protein quantitation was carried out by two independent methods:

1. UV spectroscopy at 280 nm using the absorbency value of 0.8511 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein

2. Analysis by RP-HPLC, using a standard solution of FGF-basic as a Reference Standard.
