DATA SHEET:recombinant Human Fibroblast Growth Factor-acidic

FGF-acidic is one of 23 known members of the FGF family. Proteins of this family play a central role during prenatal development and postnatal growth and regeneration of a variety of tissues, by promoting cellular proliferation and differentiation. FGF-acidic is a non-glycosylated heparin binding growth factor that is expressed in the brain, kidney, retina, smooth muscle cells, bone matrix, osteoblasts, astrocytes and endothelial cells. FGF-acidic has the ability to signal through all the FGF receptors.  This recombinant human FGF-acidic is a 15.8 kDa protein consisting of 140 amino acid residues.

Purity: >95% by SDS-Page analysis and HPLC analysis. Endotoxin less than 0.1ng per ug (1EU/ug) of FGF-a.

Reconstitution: FGF-acidic is not stable in low buffer concentration and low pH. The lyophilized powder should be reconstituted in 50ul of 10mM Tris 7.6 to a concentration of 1ug/ul. Allow to set at least 30 minutes at 4 DEG C, tighten cap, mix gently by inversion. If desired, can centrifuge 1000 X G   for 1 minute to concentrate material in vial. This solution can be diluted into other buffered solutions or stored at 4 DEG C for up to 1 week or at - 20 DEG C for futre use. Repeated freeze thaw cycles will result in some loss of activity. Recommend making single freeze thaw aliquots in carrier containing buffers. For most in-vitro applications, FGF-a exerts its biological activity in the concentration range of 1.0 to 50.0ng/ml.
Note:If this material is not to be used for coating or labeling (or other applications requiring carrier free material), the addition of 0.1% BSA to dilution buffers or dilution in sterile culture media is recommended to enhance stability and minimize absorption to vial. Ideally, maintain stock >50ug/ml.

Storage: The lyophilized powder is stable at room temperature for a few weeks, but it is best stored desiccated at -20 Deg C. Reconstituted FGF-A should be stored in working aliquots at -20 DEG C. AVOID FREQUENT FREEZE THAW CYCLES.

Reactivity: This FGF-A is fully biologically active when compared to standards. The ED50 as determined by the dose-dependent stimulation of thymidine uptake by Balb-c 3T3 cells expressing FGF receptors is <10ng/ml, corresponding to a specific activity of >100K units/mg.
