DATA SHEET:  recombinant Human Platelet Derived Growth Factor-BB

-PDGFs are disulfide-linked dimers consisting of two 12.0-13.5 kDa polypeptide chains, designated PDGF-A and PDGF-B chains. The three naturally occurring PDGFs; PDGF-AA, PDGF-BB and PDGF-AB, are potent mitogens for a variety of cell types including smooth muscle cells, connective tissue cells, bone and cartilage cells, and some blood cells. The PDGFs are stored in platelet alpha-granules and are released upon platelet activation. The PDGFs are involved in a number of biological processes, including hyperplasia, chemotaxis, embryonic neuron development, and respiratory tubule epithelial cell development. Two distinct signaling receptors used by PDGFs have been identified and named PDGFR-alpha and PDGFR-beta. PDGFR-alpha is high-affinity receptor for each of the three PDGF forms. On the other hand, PDGFR-beta interacts with only PDGF-BB and PDGF-AB. This recombinant human PDGF-BB is a 24.3 kDa disulfide-linked homodimer of two B chains (218 total amino acids).

Purity: >98% by SDS-Page analysis and FPLC analysis. Endotoxin less than 0.1ng per ug (1EU/ug) of PDGF-BB.

Reconstitution: The lyophilized material is soluble in water and most aqueous buffers. The lyophilized powder should be reconstituted in 10mM acetic acid to a concentration of 0.1-1mg/ml. Allow to set at least 30 minutes at 4 Deg C, rotate, mix well. If desired, centrifuge for 1-2 minutes at 500- 1000 rpm to concentrate material in vial .This solution can be diluted into other buffered solutions or stored at 4 DEG C for up to 1 week or at - 20DEG C for future use. For most in-vitro applications, PDGF-BB exerts its biological activity in the range of 0.2 to 10.0 ng/ml.

NOTE: If material is not to be used for labelling/coating etc, the addition of 0.1% BSA/HSA or media is recommended to enhance stability and reduce non specific absorption to vial container.

Storage: The lyophilized powder is stable at room temperature for a few weeks but it is best stored desiccated at -20 DEG C. Reconstituted PDGF-BB should be stored in working aliquots at -20DEG C. AVOID FREQUENT FREEZE THAW CYCLES.

Reactivity: The ED50 as determined by the dose-dependent stimulation of thymidine uptake by Balb-c 3T3 cells is less than 1.0 ng/ml which corresponds to a specific activity of greater than 1 million units per milligram.

Recombinant Human Platelet-derived Growth Factor BB   (rHuPDGF-BB) BULK

Description: Recombinant Human PDGF-BB produced in E.Coli is a double, non-glycosylated, polypeptide chain containing 109 amino acids and having a molecular mass of 24525.18 Dalton. rHuPDGF-BB is purified by proprietary chromatographic techniques.

Appearance: Sterile Filtered White lyophilized (freeze-dried) powder.

Endotoxin: Less than 0.1 ng/µg (IEU/µg) of rHuPDGF-BB.

Formulation: The protein was lyophilized after extensive dialysis against 2mM sodium phosphate buffer pH= 7.4±0.1.

Purity: Greater than 98.0% as determined by:

(a) Analysis by RP-HPLC.

(b) Anion-exchange FPLC.

(c) Analysis by reducing and non-reducing SDS-PAGE

Silver Stained gel.

Amino acid sequence: The sequence of the first five N-terminal amino acids was determined and was found to be Ser-Leu-Gly-Ser-Leu.

Dimers and aggregates: Less than 1% as determined by silver-stained SDS-PAGE gel analysis.

Solubility: It is recommended to reconstitute the lyophilized rHuPDGF-BB in sterile 18M-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.

Stability: Lyophilized rHuPDGF-BB although stable at room temperature for 3 weeks, should be stored desiccated below -18 C. Upon reconstitution rHuPDGF-BB should be stored at 4 C between 2-7 days and for future use below -18 C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please prevent freeze-thaw cycles.

BiologicalActivity: rHuPDGF-BB is fully biologically active when compared to standard. The ED50, calculated by the dose-dependant proliferation of murine BALB/c 3T3 indicator cells (measured by 3H-thymidine uptake) is < 1 ng/ml, corresponding to a Specific Activity of 1 MIU/mg.

Protein content: Protein quantitation was carried out by two independent methods:

1. UV spectroscopy at 280 nm using the absorbency value of 0.55 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).

2. Analysis by RP-HPLC, using a standard solution of PDGF-BB as a Reference Standard.
