DATA SHEET:recombinant Insulin Like Growth Factor I

-The IGFs are mitogenic polypeptide growth factors that stimulate the proliferation and survival of various cell types including muscle, bone, and cartilage tissue in vitro. IGFs are predominantly produced by the liver, although a variety of tissues produce the IGFs at distinctive times. The IGFs belong to the Insulin gene family, which also contains insulin and relaxin. The IGFs are similar by structure and function to insulin, but have a much higher growth-promoting activity than insulin. IGF-II expression is influenced by placenta lactogen, while IGF-I expression is regulated by growth hormone. Both IGF-I and IGF-II signal through the tyrosine kinase type I receptor (IGF-IR), but, IGF-II can also signal through the IGF-II/Mannose-6-phosphate receptor. Mature IGFs are generated by proteolytic processing of inactive precursor proteins, which contain N-terminal and C-terminal propeptide regions. This recombinant human IGF-I and IGF-II are globular proteins containing 70 and 67 amino acids, respectively, and 3 intra-molecular disulfide bonds

Purity: >97% by SDS-Page analysis and N-terminal sequence analysis. Endotoxin less than 0.1ng per ug (1EU/ug) of IGF-I.

Reconstitution: The lyophilized material is soluble in water and most aqueous buffers. The lyophilized powder should be reconstituted in sterile water to a concentration of 1 ug/ul. Allow to set at least 30 minutes at 4 DEG C, roate, mix well. If desired, centrifuge for 1-2 minutes at 500-1000 rpm to concentrate material in vial. This solution can be diluted into other buffered solutions or stored at - 20DEG C for future use. For most in-vitro applications, IGF-I exerts its biological activity in the range of 0.2 to 20.0 ng/ml.

Storage: The lyophilized powder is stable at room temperature but it is best stored desiccated below 0 Deg C. Reconstituted IGF-I should be stored in working aliquots at -20DEG C. AVOID FREQUENT FREEZE THAW CYCLES.

Reactivity: This IGF-I is fully biologically active when compared to standards. The ED50 as determined by the dose-dependent stimulation of thymidine uptake by Balb-c 3T3 cells is approx 1.0 ng/ml, corresponding to a specific activity of 1 million units per milligram.
