Human Cytokine/Growth Factor/Chemokines Sample Spec sheets (subject to change without notice):

recombinant Human Fibroblast Growth Factor-8

-Fibroblast Growth Factor-8 (FGF-8) , also known as Androgen induced growth factor (AIGF), is a heparin binding growth factor that stimulates the proliferation and activation and activation of cells that express FGF receptors. This recombinant human FGF-8 is a 22.4 kDa protein containing 193 amino acid residues.

Supplied: sterile filtered and the the protein was lyophilized from 10mM Tris pH8.0 & 75mM NaCl( no additives)

Source: E. Coli

Purity: >95% by SDS-Page and HPLC analyses. Endotoxin level is less than 0.1ng per ug of FGF-8.

Reconstitution: The lyophilized FGF-8 should be reconstituted in 5mM TRIS pH8.0 to a concentration of 0.1-1.0mg/ml). Allow to set at least 30 minutes at 4 DEG C, tighten cap and rotate to reconstitute any material on sides of vial. If desired, you can centrifuge at 1000 rpm for 1 minute to concentrate volume in bottom of vial. This solution can be diluted into water or other buffered solutions or stored at 4 DEG C for up to 1 week or at -20 DEG C for future use. For most in-vitro applications, human FGF-8 exerts its biological activity in the concentration range of 0.1 to 20.0 ng/ml.

Note:All The FGF's have a solubility problem and we are aware of it. To compensate there is an approx 20% overfill. After reconsittution, we recommend you spin down the reconstituted solution and withdraw the supernatant then discard the ppt. -for verification: You can 0.5ug on a gel vs a standard and will find the protein concentration is accurate. The A280 ext coeff. 20580 abs 0.1% (=1 g/L)   0.918

Note:If this material is not to be used for coating or labeling(or other applications requiring carrier free material), the addition of 0.1% BSA to dilution buffers or dilution in sterile culture media is recommended to enhance stability and minimize absorption to vial. Ideally, maintain concentartion <10ug/ml.

Storage: The lyophilized powder is stable at room temperature for a few weeks but it is best stored desiccated at -20 DEG C. Reconstituted human FGF-8 should be stored in working aliquots at -20 DEG C. AVOID FREQUENT FREEZE THAW CYCLES. EACh FREEZE THAW CYCLE WILL DIMINISH ACTIVITY.

Reactivity: The biological activity was determined by the does- dependent stimulation of thymidine uptake by BaF3 cells expressing FGF receptors yielding an ED50 <0.5ng/ml, corresponding to a specific activity of >1 X 10 E06 (1 million) units/mg.
